Agdia, Inc. is pleased to announce the commercial launch of a field-deployable rapid molecular test kit on its AmplifyRP® XRT platform for onsite detection of Cucumber vein yellowing virus (CVYV).

Growers can also send samples to Agdia’s Testing Services diagnostic laboratory for expanded cucurbit pathogen screening that now includes CVYV testing.

Cucumber vein yellowing virus is a plant pathogenic virus from the Ipomovirus genus that primarily causes disease in cucurbits, but can also infect several other crops, including weed species that act as natural host reservoirs. The virus has been detected primarily in the Mediterranean region, with reports from countries including Cyprus, Iran, Israel, Jordan, Portugal, Spain and Turkey. It is transmitted by the silverleaf whitefly (Bemisia tabaci) and mechanical means such as pruning.

While CVYV can infect various weed species, several cucurbit species are of primary economic concern, including cucumber (parthenocarpic cucumbers in particular), melon, squash and watermelon.

Symptoms can vary by host species but generally include:

  • Chlorosis
  • Vein clearing
  • Stunting
  • Necrosis
  • Fruit cracking or splitting

Some of these symptoms are similar in appearance to other viruses and nutrient deficiency issues, making accurate diagnosis essential to effective crop protection.

Extensive validation studies were conducted to demonstrate fitness-for-purpose of the new molecular assay developed by Agdia for the detection of CVYV:

  • Diagnostic specificity is 100% (58 out of 58 true negatives detected). No cross reactivity was observed with other pathogens tested during product validation.
  • The new assay successfully detected 119 out of 121 positive samples, resulting in a diagnostic sensitivity of 98.3%.
  • This test detects (or is predicted to detect via in silico analysis) all known isolates from various geographic regions, including Israel, Lebanon, Spain, Portugal, Jordan, Oman and Sudan.

About AmplifyRP®

Agdia’s new AmplifyRP® XRT assay for the detection of CVYV is based on recombinase polymerase amplification (RPA). This technology enables the rapid amplification and detection of nucleic acid targets (DNA or RNA) at a single operating temperature of 39–42 °C.

AmplifyRP® products achieve sensitivity and specificity comparable to PCR, with clear advantages over the lab-based technology. AmplifyRP® products eliminate the need for a nucleic acid purification step; crude sample extracts are prepared using a simple extraction buffer and tested directly. When paired with Agdia’s AmpliFire® Pro isothermal fluorometer (Figure 1, coming soon), the AmplifyRP® system is a user-friendly tool that can be implemented in the field or the lab by personnel with limited molecular diagnostic experience. Total assay time is less than 30 minutes when used with the AmpliFire® as a real-time assay.
